  • 1.  Advice needed for automation of Sanger sequencing

    Hi everyone

     

    Does anyone have experience automating Sanger sequencing? Our complete workflow from reaction setup, cycle sequencing, to reaction cleanup is currently done manually. We process approx. 40,000 samples a year and would like to automate this to reduce handling errors, improve reliability, and minimize hands-on time.

    Hi Patrick, while I can't help with automating the actual set up of the cycle sequencing reactions (we type of the sample sheet and PCR off of that), we do use a Beckman robot and magnetic beads to clean up the cycle sequencing reactions in 96-well plates.  It is super fast, reliable and the robot does a much better job than we can by doing it manually.  Hope this helps.  Jodie

    Hi Jodie, thanks for the info - it's very useful. We use XTerminator beads but magnetic bead cleanup could be a better options for us. What beads do you use? Potentially we have OpenTron and BioMek robots available to us.

    Hi Patrick, you can use ADS Sequencing Reaction Cleaning Beads from Edge Biosystems or D-Pure from Nimagen.  We have used both and they work the same.  Jodie
