  • 1.  Kawamoto Film Method

    Hello,

    I'm wondering if anyone is using Kawamoto Film Method.  I've been reading up on it but I can't find a video on the method.  If anyone has one they can share it would be fantastic.  I'm a see one, do one, teach one kind of learner.  I currently use the CryoJane which is great but I want to explore Kawamoto's method too.

    I have CryoJane that in my current cryostat.  I now need a new cryostat but most vendors won't allow you to "modify" the instrument and install the cryojane without nullifying the warranty and service options.

    Hi Nicole!

    I have used Kawamoto Film Method with some success years ago though never great. There's a group from Colorado State University in Fort Collins who suggested this paper to me during a talk at NSH: Full article: The use of Commercially Available Adhesive Tapes to Preserve Cartilage and Bone Tissue Integrity During Cryosectioning.

    I believe they are having some good success in their lab with tape transfer methods; you could contact Cecily Broomfield from that Colorado group through the NSH directory and see if she has some suggestions.

    Hi Nicole,

    We don't use tape transfer in our lab but I thought the following might be useful for you:

    1. I can't find a video tutorial for but I did find a detailed protocol with diagrams from the authors who developed the technique (see PDF attachment here). It seems very similar to the CryoJane. 

    1. There's also this protocol I found which is a modification of the CryoJane for mineralized specimens:
